چکیده

Overexpression of several different Aspergillus niduluns developmental regulatory genes has been shown to cause inappropriate developmental activation and growth inhibition. We previously exploited this observation that induced development caused growth inhibition in designing a screen to identify other genes that could activate development when overexpressed. We identified 16 mutants in which induced expression of different random genomic DNA sequences caused growth inhibition, accumulation of mRNA corresponding to the brk4 developmental regulatory locus, and in several cases sporulation. This phenotype was designated FAB for Forced expression Activation of brD1. and the genes were called fabA through fabP. Here we describe one of these genes, fabM, which is predicted to encode a poly(A)binding protein (PABP) that is constitutively expressed and is essential for viability. While it is unclear why overexpression of the fabM caused sporulation, we showed that this activity required other known early developmental regulators including JbA, JbB, JbC, and JUG. We propose that fabM is an example of a gene that is not only required for growth, but also has specific functions early in development that assist developmental induction, presumably by allowing translation of specific mRNAs like

Refolding Process of Cysteine-Rich Proteins: Chitinase as a Model

Background: Recombinant proteins overexpressed in E. coli are usually deposited in inclusion bodies. Cysteines in the protein contribute to this process. Inter- and intra- molecular disulfide bonds in chitinase, a cysteine-rich protein, cause aggregation when the recombinant protein is overexpressed in E. coli. Hence, aggregated proteins should be solubilized and allowed to refold to obtain nat...

tudy on sex steroid-binding proteins (with emphasize on 17 -estradiol) in plasma of female and juvenile kutum (Rutilus frisii kutum)

A sex steroid-binding protein (SBP) that binds to 17 b-estradiol with high affinity and moderate capacity was identified in the plasma of female and juvenile of kutum (Rutilus frisii kutum) sampled during the early stage of gonadal maturation in October and prior to spawning in March. Mean maximum specific binding (Bmax) and equilibrium dissociation constant (Kd) of the fish were as follows: In...

IMPROVEMENT OF hGM-CSF EXPRESSION USING A FUSION SYSTEM BY DIRECTING PROTEIN TO THE PERIPLASMIC SPACE

Bacterial expression systems provide economic and logistic advantages in manufacturing proteins for human therapeutic purposes. However, most such proteins accumulate in insoluble biologicaIIy inactive form when overexpressed in bacterial cells. This is the case while attempting to produce recombinant human granulocyte-macrophage colony stimulating factor (hGM-CSF) in E. coli. In this repor...

Roles of the C terminus of Armadillo in Wingless signaling in Drosophila.

Drosophila melanogaster Armadillo and its vertebrate homolog beta-catenin play multiple roles during development. Both are components of cell-cell adherens junctions and both transduce Wingless (Wg)/Wnt intercellular signals. The current model for Wingless signaling proposes that Armadillo binds the DNA-binding protein dTCF, forming a bipartite transcription factor that activates Wingless-respo...

Differential protein expression in Mycobacterium tuberculosis susceptible and multidrug resistant isolates

Introduction: Infections by multidrug resistant Mycobacterium tuberculosis (MDR-TB) is a major public health challenge. Secretion of proteins by M. tuberculosis plays an important role in the pathogenesis of the bacterium. We compared the protein profiles of susceptible M. tuberculosis and MDR-TB isolates using proteomic analyses, namely two dimensional gel electrophoresis (2DE) and mass spectr...
